A new nasal spray that offers protection from all COVID variants has successfully passed lab tests and will now advance to human trials.
Since the virus has continued to evolve over the years, early treatments, including vaccines, have become less effective at combating the new variants.
Last month, several antibody treatments — which require complex refrigerated supply chains — were halted after failing to offer protection against the BA.2 omicron sub-variant.
Meanwhile, the new spray is a much simpler solution. It contains “potent” proteins which target the virus’s weak spots and prevent it from infecting the body, according to researchers at Northwestern University and the University of Washington.
Tests on mice showed promising results, which were published in the journal Science Translational Medicine on April 12, 2022. Other nasal sprays are also being tested in England and Israel.
The American research team started by using supercomputers to design proteins that could stick to the virus’s vulnerable spots, such as the spike protein.
They then re-engineered the proteins, called minibinders, to make them even more potent. These potent minibinders now simultaneously bind to three sites of the virus instead of just one, making it less likely to detach.
Common antibody therapies may only block one site, but SARS-CoV-2’s spike protein has three binding domains, explained Professor Michael Jewett from the University of Washington School of Medicine.
“Our minibinders sit on top of the spike protein like a tripod and block all three,” he said. “The interaction between the spike protein and our antiviral is among the tightest interactions known in biology. When we put the spike protein and our antiviral therapeutic in a test tube together for a week, they stayed connected and never fell apart.”

The proteins were able to prevent COVID from binding to the ACE2 receptor, the entry point for infecting the body. If the virus is unable to bind to the receptor, it cannot infect the body, suggesting that the treatment will work against new variants.
The treatment is a more accessible alternative to vaccines, which require a healthcare professional to administer. There is a high chance that it can be self-administered via nasal spray.
They can also be produced on a large scale since they are far more cost-effective to manufacture and are stable in high heat.
The researchers hope that the human trials will show similar success so that the treatment can be made available in pharmacies.
